Target Information
Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway, is crucial for normal growth control and may be defective in tumor cells. It preserves cellular nutrients under starvation and maintains cytosolic turnover.
Beclin-1, a key regulator of autophagosome formation, is regulated by Ambra1. Ambra1 associates with Beclin-1 near its center, as shown by yeast two-hybrid assay.
Null mutations of Ambra1 in mice lead to embryonic lethality with severe neural tube defects due to impaired autophagy, accumulation of ubiquitinated proteins, and excessive apoptosis.
Down-regulation of Ambra1 via RNA interference reduces rapamycin- and starvation-induced autophagy. Multiple isoforms of Ambra1 are known to exist.
