Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
  • For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (~90 kDa) has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

AR signaling is essential for the development and function of male reproductive organs (prostate, epididymis) and also plays roles in nonreproductive tissues (muscle, hair follicles, brain). AR is a phosphoprotein that regulates MAP kinase signaling; inhibition of the MEK1/2 pathway alters its phosphorylation state. Dysregulation of AR signaling is linked to diseases such as prostate cancer, Kennedy’s disease, and male infertility. Mutations in AR are associated with complete androgen insensitivity syndrome (CAIS).
