Target Information
The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (~90 kDa) contains three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain.
AR is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
AR signaling is essential for the development and function of male reproductive organs (prostate, epididymis) and also influences nonreproductive tissues such as muscle, hair follicles, and brain.
As a phosphoprotein, AR regulates the MAP kinase pathway, and MEK1/2 inhibition alters its phosphorylation state.
Dysregulation of AR signaling is associated with prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene can lead to complete androgen insensitivity syndrome (CAIS).
