Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 160 kDa surface receptor from the EGF-TM7 family. It contains seven EGF-like domains, seven transmembrane sequences, and an intracellular C-terminal domain homologous to other TM7 superfamily members.
F4/80 is a well-established marker for mature macrophages and is also expressed by peritoneal eosinophils, Langerhans cells, and some dendritic cell subsets. Expression begins during early myeloid development and can be upregulated by M-CSF stimulation. Some macrophage populations, particularly in lymphoid tissue, may lack F4/80 expression.

Brilliant Ultra Violet™ 661 (BUV661) is a tandem dye emitting at 660 nm, suitable for cytometers with a 355 nm UV laser.
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) with minimal impact on brightness and FRET efficiency.

Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein, part of the EGF TM7 family, sharing 68% identity with human EMR1.
It is expressed on diverse mature macrophage populations, including Kupffer cells, Langerhans cells, microglia, and macrophages in the spleen, thymus, lung, and bone marrow.
Expression varies with macrophage maturation and activation. F4/80 is also found on some dendritic cells but absent from macrophages in T cell regions of lymphoid organs.
Its biological functions are not fully defined, though studies suggest a role in CD8⁺ regulatory T cell generation.
