Product Specific Information

This antibody detects endogenous levels of PKR only when phosphorylated at Threonine 446.

Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses. EIF2AK2 plays a key role in antiviral defense mediated through its direct activation by double-stranded RNA produced during viral infection. PKR is involved in IFN-induced innate antiviral response, virus-induced apoptosis, and regulation of cell growth and differentiation. It induces apoptosis via Fas expression and mediates the FADD/Caspase-8 death signaling pathway. Upon viral infection, PKR can activate both cell survival and cell death pathways through kinase-independent and -dependent mechanisms. PKR also regulates NF-kappaB activation, p53, p38, and PDGF signaling pathways, and has been implicated in tumor suppression, malignancy, and several neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses have evolved mechanisms to evade PKR’s antiviral effects, such as inhibition by the influenza virus NS1 protein.
