Product Specific Information

This 1E8 monoclonal antibody reacts with human alpha-fetoprotein (AFP), a 70-kDa secretory protein belonging to the albumin gene family. AFP is synthesized by the yolk sac and fetal liver during embryogenesis, with high levels in fetal serum. After birth, AFP levels decrease significantly and are nearly undetectable in normal adults. Elevated AFP levels are associated with hepatocellular carcinoma, germ cell teratoblastoma, liver regeneration, viral hepatitis, and cirrhosis. Therefore, AFP detection is used as a diagnostic marker for these conditions.

Applications Reported: Suitable for microscopy, immunohistochemical staining, and immunocytochemistry.
Applications Tested: Tested by immunohistochemistry on formalin-fixed paraffin-embedded tissue using high pH antigen retrieval at 20 µg/mL. Optimal titration is recommended for best performance.

Target Information

AFP (Alpha Fetoprotein) is a major plasma protein produced by the yolk sac, intestinal tract, and liver during fetal life. In adults, AFP expression is often associated with hepatoma or teratoma, though hereditary persistence may occur without pathology. AFP is considered the fetal counterpart of serum albumin, with both genes located tandemly on chromosome 4. AFP exists in monomeric, dimeric, and trimeric forms and can bind copper, nickel, fatty acids, and bilirubin. AFP levels in amniotic fluid are used to detect neural tube defects such as spina bifida and anencephaly. Elevated serum AFP is observed in hepatocellular carcinoma, teratoblastoma, colorectal, pancreatic, and germ cell cancers.
