Specificity Comments:
Receptor tyrosine kinases (RTKs) are transmembrane molecular scaffolds that regulate key cellular processes such as cell cycle, migration, metabolism, and survival. IGF1R (Insulin-like Growth Factor-I Receptor) is an RTK that stimulates growth, inhibits apoptosis, and mediates growth hormone responses. IGF-I binding promotes association of IGF1R with Shc, GRB2, and Sos1, initiating Ras and ERK kinase cascades that activate transcription factors like Elk. IRS-1 and IRS-2 PTB domains interact with active IGF1R to trigger PI3K-dependent downstream signaling.
The human IGF1R gene is located on chromosome 15q26.3 and encodes a 1,376 amino acid precursor protein, while IGF2R is located on chromosome 6q26 and encodes a 2,491 amino acid transmembrane protein.

Target Information

IGF1R (Insulin-like Growth Factor 1 Receptor) is a receptor tyrosine kinase activated by IGF-1 and IGF-2 and is implicated in various cancers. The protein forms a tetramer of two α and two β subunits. The α-subunit binds IGF-1 with higher affinity than IGF-2, leading to conformational change and autophosphorylation at Tyr1134, Tyr1135, and Tyr1136. Activated IGF1R triggers phosphorylation cascades via RAS, RAF, and MAPK pathways (ERK, p38, JNK), resulting in transcription of genes promoting proliferation.
