Target Information
IGF1R, also known as Insulin-like Growth Factor 1 (IGF-1) Receptor, is a receptor tyrosine kinase activated by IGF-1 and IGF-2. IGF1R has been implicated in a variety of cancers. The protein forms a tetramer consisting of two alpha and two beta subunits. The alpha-subunits bind to IGF-1 with higher affinity than IGF-2. Binding induces a conformational change, followed by autophosphorylation of IGF-1R at positions 1134, 1135, and 1136. The activated receptor phosphorylates substrate proteins, leading to sequential activation of RAS, RAF, and MAPK isoforms (ERK, p38, JNK), which drive gene transcription and cell proliferation.
