Product Specific Information
Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes.
It is absent on non-hematopoietic cells and tissues. CD45 glycoprotein plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases and cytokine receptor signaling.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
CD45 is essential for T- and B-cell receptor signaling, integrin-mediated pathways, apoptosis regulation, and various immunological functions.
