- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD45 (LCA, leukocyte common antigen)은 모든 핵을 가진 조혈 세포에 보편적으로 발현되는 수용체형 단백질 티로신 포스파테이스(PTP)입니다. 림프구 표면 단백질의 약 10%를 차지하며, 비조혈 세포나 비조혈 조직에서는 발현되지 않습니다.
CD45는 림프구 발달과 항원 신호 전달에 중요한 역할을 하며 Src-family kinase의 조절 인자로 작용합니다.
대체 스플라이싱으로 인해 여러 동형 단백질(isoform)이 존재하며, 세포 유형과 생리적 상태에 따라 발현이 달라집니다.
CD45는 T세포 및 B세포 항원 수용체 신호 조절, JAK kinase 억제를 통한 사이토카인 수용체 신호 조절, 세포 생존 및 면역 기능 조절에 핵심적인 역할을 합니다.
