• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) ubiquitously expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes.
It is absent on non-hematopoietic cell lines and tissues.
CD45 glycoprotein plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
CD45RA is an isoform with restricted expression among lymphoid cell subtypes.
CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases, and modulates cytokine receptor signaling.
It also promotes cell survival by influencing integrin-mediated signal transduction, DNA fragmentation during apoptosis, and various immunological functions.
