Target Information
Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by several aggregation-prone proteins and maintains unfolded proteins for refolding by other chaperones. Clusterin does not refold proteins independently but mediates their internalization and degradation through lysosomal or proteasomal pathways. Secreted isoforms protect cells from apoptosis and complement-mediated cytolysis, while intracellular and nuclear isoforms regulate ubiquitination, NF-κB activity, and apoptosis. Mitochondrial isoforms suppress BAX-dependent cytochrome c release, contributing to cell survival and proliferation regulation.
