Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins. It inhibits amyloid fibril formation by various aggregation-prone proteins and maintains partially unfolded proteins for subsequent refolding by other chaperones such as HSPA8/HSC70. Secreted isoform 1 protects cells against apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ligase complexes, promoting proteasomal degradation of target proteins and modulating NF-kappa-B activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis, playing a role in cell proliferation regulation.
