Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease. It regulates various physiological processes including:
- Cell survival, migration, and differentiation
- Phagocytosis of apoptotic cells (efferocytosis)
- Autophosphorylation upon ligand binding, providing docking sites for downstream signaling molecules
Following activation, MERTK interacts with GRB2 or PLCG2 and induces phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and engulfment. In RPE, it regulates rod outer segment fragment phagocytosis and inhibits Toll-like receptor (TLR)-mediated innate immune responses by activating STAT1, which induces SOCS1 and SOCS3 production.
