Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human sCD34. Anti-Human sCD34-specific antibody was purified by affinity chromatography employing an immobilized Human sCD34 matrix.

Sandwich ELISA:
To detect Human sCD34 by sandwich ELISA (using 100 µL/well), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human sCD34 (500-P327) as a capture antibody, allows the detection of at least 2000–4000 pg/mL of Recombinant Human sCD34.

Western Blot:
To detect Human sCD34 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible development reagents, the detection limit for Recombinant Human sCD34 is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) present on many stem cell populations. It serves as a stem cell marker, although its expression on human hematopoietic stem cells is reversible. CD34 may act as a receptor mediating endocytosis and regulating adhesion, differentiation, and proliferation of hematopoietic stem cells and progenitors. It likely plays a role in stem cell attachment to the bone marrow extracellular matrix or stromal cells and may act as a scaffold for glycans binding to lectins. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
