Target Information

Tight junctions are specialized regions of cell-cell contact abundant in luminal epithelial sheets. In electron micrographs, they appear as belt-like sealing strands encircling each cell, forming a molecular gasket that prevents leakage of ions and molecules.
Beyond their barrier function, tight junctions also maintain epithelial polarity by blocking diffusion of membrane proteins between apical and basolateral surfaces.
Proteins such as ZO-1, ZO-2, ZO-3, cingulin, Rab-3b, and symplekin are associated with tight junctions, contributing to assembly, maintenance, and permeability regulation.
Claudin-1 and Claudin-2 are key structural components of tight junction strands, unlike occludin which plays an accessory role. Claudin-2 functions as a paracellular channel selective for cations (Na+) and is mainly expressed in liver and kidney, with minor expression in brain.
