Target Information
Tight junctions are specialized regions of cell-cell contact abundant in luminal epithelial cell sheets. In freeze-fracture electron micrographs, tight junctions appear as belt-like bands of sealing strands encircling each cell. These strands form a molecular gasket preventing ions and molecules from leaking between cells.
Tight junctions also maintain epithelial polarity by blocking diffusion of membrane proteins between apical and basolateral surfaces. Proteins such as ZO-1, ZO-2, ZO-3, cingulin, Rab-3b, and symplekin are associated with tight junctions, contributing to assembly, maintenance, and permeability regulation.
Claudin-1 and Claudin-2 are major structural components of tight junction strands, inducing networks typical of true tight junctions. Claudin-2 functions as a paracellular channel selective for cations (Na+) and is primarily expressed in liver and kidney, with minor expression in the brain.
