Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II, associated with BCR signaling, inducing tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; recommended ≤0.2 µg/test (100 µL final volume). Optimize antibody titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before mixing with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
• NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescence with minimized spillover and spread.
• Excitation: 552 nm / Emission: 663 nm / Laser: 561 nm (Yellow).
• Non-specific B cell staining observed in SJL mice; not in BALB/c or C57BL/6.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains, expressed on nearly all peripheral B cells. It is also found on certain myeloid leukemia cells. CD19 functions as a co-receptor with BCR, amplifying antigen receptor signaling, and plays a critical role in B cell activation and regulation. Mutations in CD19 can lead to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
