Product Specific Information

The eBio1D3 (1D3) monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed on B cells during all developmental stages except terminal plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, and MHC class II, associating with the BCR to mediate signaling, tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications

• Reported for use in flow cytometric analysis
• Tested by flow cytometric analysis of mouse splenocytes
  Recommended ≤0.2 µg per test (100 µL final volume)

Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as PI, 7-AAD, or DAPI.
  Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• NovaFluor conjugates require CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  Add 5 µL per sample prior to combining with cells.
• Excitation: 552 nm; Emission: 592 nm; Laser: 561 nm (Yellow)
• Internal testing shows NovaFluor Yellow 590 may non-specifically stain B cells in SJL mice.
  No non-specific staining observed in BALB/c or C57BL/6 strains.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on nearly all peripheral B cells and certain myeloid leukemia cells. CD19 functions as a signal-amplifying coreceptor for the B cell receptor (BCR) and regulates normal and malignant B cell proliferation. Mutations in CD19 can cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
