Product Specific Information

The M1/69 monoclonal antibody reacts with the mouse CD24 molecule (Heat Stable Antigen, HSA).
CD24 is a 35–50 kDa GPI-anchored, variably glycosylated membrane molecule expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells.
It is used to resolve stages of B lymphopoiesis in mouse bone marrow and may bind to P-selectin (CD62P).

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes and thymocytes.
Recommended use ≤ 0.8 µg per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton™ nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread impacts.

Excitation: 552 nm
Emission: 592 nm
Laser: 561 nm (Yellow)

Note: NovaFluor Yellow 590 may non-specifically stain B cells in SJL mice.
No non-specific staining observed in BALB/c or C57BL/6 strains.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B cells to mature B cells.
Expression is low or absent in plasma cells.
CD24 is also found in B-lineage acute lymphoblastic leukemias, B-cell CCLs, and non-Hodgkin’s lymphomas.
It may regulate B-cell proliferation, maturation, and autoimmunity.
