Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-21. Anti-Human IL-21-specific antibody was purified by affinity chromatography employing an immobilized Human IL-21 matrix.

Sandwich ELISA:
For detection of Human IL-21 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. In conjunction with PeproTech Biotinylated Anti-Human IL-21 (500-P191Bt) as a detection antibody, it enables detection of at least 0.2–0.4 ng/well of Recombinant Human IL-21.

Western Blot:
For detection of Human IL-21 by Western blot analysis, use at 0.1–0.2 µg/mL. With compatible secondary reagents, the detection limit for Recombinant Human IL-21 is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer (NKT), T helper (Th17), and T follicular helper (TFH) cells.
In TFH cells, IL-21 induces autocrine signaling through IL-21R and STAT3, leading to transcriptional activation by Bcl6. Similar to IFN-γ for Th1 and IL-4 for Th2, IL-21 is essential for TFH cell development and function.
IL-21 promotes T cell-dependent B cell differentiation into plasma and memory cells, stimulates IgG production, and induces apoptosis in naive B cells.
In Th17 cells, IL-21 enhances IL-23R expression via STAT3, IRF4, and RORγt, supporting Th17 maturation and maintenance. It downregulates Foxp3 expression and is linked to colitis models, where IL-21 deficiency confers protection.
