Product Specific Information

The 14-4-4S monoclonal antibody reacts with mouse and rat MHC class II, I-E subregion-encoded glycoprotein in strains of H-2^k, H-2^d, H-2^p, and H-2^r. Haplotypes H-2^b, H-2^f, and H-2^s do not express the I-E antigen.
MHC class II is present on B cells, monocytes, macrophages, and dendritic cells. The 14-4-4S mAb has been reported to block antigen presentation in vitro and cross-reacts with rat.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes, ≤0.06 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Careful titration is recommended for optimal performance.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells (macrophages, dendritic cells, B cells). They bind exogenous antigens in endosomes before being displayed on the cell surface for T-cell recognition.
The antigen-binding groove between the alpha and beta chains is open at both ends and accommodates peptides 15–24 amino acids long.

HLA and MHC antibodies are valuable tools in Immunopeptidomics, aiding in the identification and characterization of neoantigens via LC-MS/MS.
