Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV; SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of human peripheral blood cells (5 µL/test; 0.1 µg/test, 100 µL final volume).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), required for optimal staining. Add 5 µL per sample or per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology, providing engineered fluorescence with optimized spillover and spread characteristics.

Target Information

CD4 antigen is a co-receptor for HIV and recognizes MHC class II molecules. Expressed mainly in T helper cells, monocytes, macrophages, and dendritic cells. Found in thymus, lymph nodes, tonsils, spleen, and certain brain and gut tissues. CD4 assists in T-cell activation via association with TCR and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases. Multiple splice variants identified (RefSeq, July 2017).
