Product Specific Information
This TNKUPJ monoclonal antibody recognizes mouse CD163, a 130 kDa surface receptor expressed by specific tissue macrophage subsets such as splenic red pulp macrophages, Kupffer cells, intestinal lamina propria macrophages, and a small fraction of peritoneal macrophages. Mouse monocytes do not express CD163, and unlike human CD163, it is not effectively induced by M2 polarizing cytokines.
The antibody detects CD163 on fixed and permeabilized cells, enabling staining of intracellular pools. Aggressive tissue dissociation may reduce surface CD163; intracellular detection is recommended in such cases.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell count may vary (10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.
Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm, suitable for cytometers with UV (355 nm) laser. Protect from light due to photo-induced oxidation sensitivity.
Buffer Recommendations:
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific polymer interactions when multiple polymer dye-conjugated antibodies are used.
Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) with minimal brightness loss.

Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, part of the scavenger receptor cysteine-rich superfamily and receptor for hemoglobin-haptoglobin complexes. It protects tissues from oxidative damage and is involved in iron recycling through endocytosis of hemoglobin/haptoglobin and heme breakdown.
CD163 is expressed on human monocytes and macrophages, upregulated by IL-10, IL-6, and dexamethasone, and shed upon LPS or PMA stimulation. It binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, triggering intracellular signaling cascades leading to IL6 and CSF1 secretion.
