Product Specific Information

The 6H6 monoclonal antibody reacts with human CD123, the alpha chain of the IL-3 receptor. This 60–70 kDa transmembrane protein binds IL-3 with low affinity by itself, and with CD131 (common beta chain) forms a high-affinity receptor. CD123 is expressed by myeloid precursors, macrophages, dendritic cells, mast cells, basophils, and megakaryocytes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and is intended for use on cytometers equipped with a UV (355 nm) laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal fluorescence loss.

It transmits IL-3 signals and is overexpressed on acute myeloid leukemia (AML) blasts compared to normal hematopoietic cells, representing a potential therapeutic target.
