Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Murine IL-21. Anti-Murine IL-21-specific antibody was purified by affinity chromatography employing an immobilized Murine IL-21 matrix.

Sandwich ELISA:
To detect Murine IL-21 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Murine IL-21 (500-P278Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Murine IL-21.

Western Blot:
To detect Murine IL-21 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. Used in conjunction with compatible secondary reagents, the detection limit for Recombinant Murine IL-21 is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer Cells (NKT), T helper (Th17), and T follicular helper (TFH) cells.
In TFH cells, IL-21 expression leads to autocrine signaling through the IL-21 receptor (IL-21R) and STAT3, which activates transcription via Bcl6.
IL-21 is critical for TFH cell development and effector function, promoting B cell differentiation into plasma and memory cells, stimulating IgG production, and inducing apoptotic signaling in naive B cells.
In Th17 cells, IL-21 upregulates IL-23R through STAT3, IRF4, and RORγt, preparing Th17 cells for maturation and maintenance by IL-23.
IL-21 also downregulates Foxp3, and IL-21-deficient mice are protected from chemically induced colitis due to reduced Th17-associated molecule expression. Elevated IL-21 levels are found in colitis mouse models.
