Product Specific Information

The monoclonal antibody 51.1 reacts with human CD1d, a member of the CD1 family with similarity to non-polymorphic MHC Class I-like molecules. CD1d is a conserved transmembrane receptor of the Immunoglobulin Superfamily, capable of associating with beta-2 microglobulin or existing independently as a nonglycosylated protein. This dual expression suggests different mechanisms for presenting glycolipid molecules to CD1d-reactive NKT cells.

CD1d is expressed on peripheral B cells, resting monocytes, cortical thymocytes, and polarized on intestinal epithelial cells. Low-level intracellular expression is observed in hepatocytes, and elevated expression is reported in HCV-infected livers.

The 51.1 monoclonal antibody has functional activity, capable of blocking interactions between CD1d-transfected cells and NKT cells.

Applications Reported: Flow cytometry, immunoprecipitation, and immunohistology of frozen tissue sections.
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.5 µg/test (in 100 µL final volume). Cell count: 10⁵–10⁸ cells/test. Optimize by titration for best performance.

Target Information

CD1d is a heavy chain associated with beta-2 microglobulin on cortical thymocytes, and can also be expressed independently on intestinal epithelial cells. It is the sole group 2 member of the CD1 family, encoding a transmembrane glycoprotein structurally related to MHC proteins that forms heterodimers with beta-2-microglobulin.

CD1 proteins mediate presentation of lipid and glycolipid antigens to T cells. The human genome contains five CD1 family genes located on chromosome 1, differing in localization and lipid ligand specificity. CD1d localizes to late endosomes and lysosomes through a tyrosine-based motif in its cytoplasmic tail.
