Product Specific Information

The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein expressed on most acute lymphoblastic leukemias (ALL). CD10 is identical to neutral endopeptidase (NEP) and functions as a Zn²⁺-dependent metallo-peptidase involved in chemotactic and inflammatory regulation.

Major expression sites include:

• B cells (stromal cell-dependent lymphopoiesis)
• Germinal center B cells
• Brush border of enterocytes
• Renal tubules and glomeruli

Partial blocking occurs between eBioCB-CALLA and MEM-78 antibodies, indicating similar epitope recognition.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells. Use 5 µL (0.2 µg)/test per 100 µL sample (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining and should be added before combining antibodies with cells (5 µL per sample).
• For single-color controls, use 5 µL of CellBlox Blocking Buffer per 100 µL cell sample.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface neutral metalloendopeptidase that inactivates various biologically active peptides. It is expressed in:

• Lymphoblastic, Burkitt’s, and follicular germinal center lymphomas
• Immature B cells and chronic myelocytic leukemia (CML) cells
• Breast myoepithelial cells, bile canaliculi, fibroblasts
• Kidney and gut epithelial brush border

CD10 cleaves peptides at the amino side of hydrophobic residues and inactivates peptide hormones such as glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. The CD10 gene produces four mRNA transcripts via alternative splicing (coding region unaffected).
