Product Specific Information

The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified in acute lymphoblastic leukemia (ALL). CD10 functions as a Zn²⁺-dependent metallo-peptidase involved in regulating chemotactic and inflammatory processes. It is expressed in B cells, enterocytes, renal tubules, and glomeruli.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

• Flow cytometric analysis.

Applications Tested

• Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
• Recommended usage: 5 µL (0.2 µg) per test (100 µL final volume).
• Suitable for 10⁵–10⁸ cells/test.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer

• Updated formulation (Cat. No. C001T06F01) for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD10 (CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides.
It is expressed in lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, and certain epithelial tissues such as kidney and gut brush borders.
CD10 cleaves peptides at the amino side of hydrophobic residues and inactivates hormones like glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin.
The CD10 gene produces multiple mRNA transcripts due to alternative splicing in the 5′ untranslated region.
