Target Information
Two distinct signaling pathways activate the host innate immunity against viral infection. One pathway relies on members of the Toll-like receptor (TLR) family, while the other uses the RNA helicase RIG-I as a receptor for intracellular viral double-stranded RNA to trigger the immune response.
MAVS is a mitochondrial membrane protein critical in IFN beta signaling, recruiting IRF-3 to RIG-I and activating NF-kappa-B. It interacts with innate immunity components such as TRIF, TRAF2, and TRAF6, and also with kinases IKK-alpha, IKK-beta, and IKK-iota through its C-terminal region. Cleavage of this region by the Hepatitis C virus protease allows viral immune evasion. Multiple isoforms of MAVS are known.
