Target Information
Janus kinase 2 (JAK2)는 특정 사이토카인 수용체 신호 전달 경로에 관여하는 단백질 티로신 키나아제로, polycythemia vera에서 변이가 보고되어 있습니다.
JAK2는 JH1 촉매 도메인을 포함하며, 활성화된 JAK은 STAT 단백질을 인산화시켜 핵으로 이동하게 합니다.
핵 내에서 STAT 단백질은 다양한 유전자의 전사를 조절하며, JAK2는 IFN-γ 수용체 복합체의 신호 개시에 필요합니다.
JAK1은 증폭자로 작용하며, 일부 연구에서는 Tyk2 및 JAK3가 IFN-γ 수용체 복합체 내에서 적절히 위치할 경우 JAK2의 역할을 대체할 수 있다고 제안합니다.
JAK2 변이는 Budd-Chiari 증후군, Polycythemia vera, Thrombocythemia 3, Myelofibrosis, 또는 Acute myelogenous leukemia를 유발할 수 있습니다.
