Product Specific Information

The B20.1 monoclonal antibody reacts with mouse T-cell receptor (TCR) V alpha 2 chains, recognizing the majority of the TCR V alpha 2 subfamily in mice carrying the a, b, and c haplotypes. It also reacts with TCR V delta 8 products due to high homology.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Flow cytometry of mouse lymph node cells (≤0.1 µg/test, 100 µL final volume)

Recommended Usage:

• Preblock Fc receptors with 0.5 µg of purified anti-mouse CD16/32 (Product #14-0161-82) for 10 minutes.
• Titrate antibody for optimal performance.
• Cell number: 10⁵–10⁸ cells/test (empirically determined).

NovaFluor Dyes Compatibility:

• Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer:

• Product #C001T06F01
• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample (or per 100 µL for single-color controls).

Target Information

T cell receptors (TCR) are essential for adaptive immune responses, recognizing foreign versus self-peptides presented by MHC class II molecules. Abnormal TCR recognition is linked to autoimmune diseases and tumor-associated mutations. Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies to TCR V beta segments are associated with rheumatoid arthritis and systemic lupus erythematosus, potentially modulating immune responses. Most human T cells express TCR alpha-beta with CD4 or CD8, while gamma-delta T cells, found mainly in epithelia, exhibit distinct antigen recognition and immunoregulatory activity.
