Product Specific Information

The B20.1 monoclonal antibody reacts with mouse T-cell receptor (TCR) V alpha 2 chains. It recognizes the majority of the TCR V alpha 2 subfamily in mice carrying the a, b, and c haplotypes, and also reacts with V delta 8 products due to high homology.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis

Applications Tested:

• Tested by flow cytometric analysis of mouse lymph node cells
• Recommended usage ≤ 0.1 µg per test (100 µL final volume)
• Cell number range: 10^5–10^8 cells/test
• Preblock Fc receptors for 10 min using 0.5 µg purified anti-mouse CD16/32 (Product #14-0161-82)
• Titrate antibody for optimal performance

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment

CellBlox Plus Blocking Buffer Guidance:

• Updated conjugate ships with CellBlox Plus Blocking Buffer (Product #C001T06F01)
• Add 5 µL per sample to antibody cocktails before combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10^3–10^8 cells)

TCR recognition of self-peptides is linked to autoimmune diseases and tumor-associated mutations.
Superantigen engagement can lead to toxic shock syndrome.
Autoantibodies to TCR V beta segments are found in RA and SLE patients, potentially modulating immune responses.
Most human T cells express TCR alpha-beta and either CD4 or CD8; a small subset (double negative) lacks both.
Gamma-delta T cells, primarily epithelial, exhibit limited diversity and unique antigen recognition, with antitumor and immunoregulatory roles.
