Product Specific Information

The B20.1 monoclonal antibody reacts with mouse T-cell receptor (TCR) V alpha 2 chains, recognizing most of the TCR V alpha 2 subfamily in mice with a, b, and c haplotypes. It also cross-reacts with TCR V delta 8 due to high homology.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested with mouse lymph node cells at ≤0.1 µg/test (100 µL final volume).

Recommended Protocol:

• Preblock Fc receptors for 10 min using 0.5 µg purified anti-mouse CD16/32 (Product #14-0161-82).
• Titrate antibody for optimal performance.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains; avoid DNA intercalating dyes (PI, 7-AAD, DAPI).

CellBlox Plus Blocking Buffer (Product #C001T06F01):

• Improved formulation for NovaFluor conjugates.
• Add 5 µL per sample before mixing with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Dyes:

• Based on Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.
• Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).

Note: NovaFluor Yellow 610 may stain B cells non-specifically in SJL mice; not observed in BALB/c or C57BL/6 strains.

Target Information

T-cell receptors (TCRs) enable discrimination of foreign vs. self-peptides presented by MHC class II molecules, critical for adaptive immunity.
Abnormal TCR recognition is associated with autoimmune diseases and tumors.
Superantigen engagement of TCRs can lead to toxic shock syndrome.
Autoantibodies to TCR V beta segments are found in RA and SLE patients, potentially modulating inflammatory responses.
Most human T cells express TCR alpha-beta with CD4 or CD8, while gamma-delta T cells exhibit distinct antigen recognition and immunoregulatory functions.
