Target Information

Presenilin1 was initially identified as a marker of susceptibility to early-onset Alzheimer’s disease. Together with PEN2, nicastrin, and APH-1, Presenilin1 forms the γ-secretase protein complex—a membrane-bound aspartyl protease that cleaves certain proteins at peptide bonds buried within the lipid bilayer. This cleavage is essential in signaling from several cell-surface receptors and is involved in generating neurotoxic amyloid peptides central to Alzheimer’s disease pathogenesis.
γ-secretase cleaves amyloid precursor protein (APP) within its intramembrane region, producing either non-toxic p3 (α/γ cleavage) or toxic Aβ amyloid peptide (β/γ cleavage). Accumulation of Aβ peptide is considered a precursor to Alzheimer’s disease. Multiple isoforms of Presenilin1 exist.
