Target Information
Replication factor C (RFC) is an essential DNA polymerase accessory protein required for various aspects of DNA metabolism, including DNA replication, repair, and telomere maintenance. RFC functions as a heteropentameric complex that recognizes primer-template DNA, binds to primer termini, and loads proliferating cell nuclear antigen (PCNA) onto DNA in an ATP-dependent manner. All RFC subunits share conserved RFC box sequences containing nucleotide-binding motifs. Four RFC genes (RFC1–RFC4) possess ATP-binding motifs. Smaller RFC subunits (RFC2–RFC5) interact with Rad24, whereas RFC1 does not. RFC2, the third-largest subunit, plays a crucial role in ATP binding, DNA replication, and checkpoint functions.
