Target Information
Replication factor C (RFC) is an essential DNA polymerase accessory protein required for DNA replication, repair, and telomere metabolism. RFC is a heteropentameric complex that recognizes primer-template DNA, binds to the primer terminus, and loads proliferating cell nuclear antigen (PCNA) onto DNA in an ATP-dependent manner. All five RFC subunits share conserved RFC boxes containing nucleotide-binding motifs. RFC1–RFC4 possess ATP-binding motifs, while smaller subunits (RFC2–RFC5) interact with Rad24, unlike RFC1. RFC2, the third-largest subunit, exhibits ATP binding crucial for DNA replication and checkpoint function.
