Product Specific Information

The monoclonal antibody XLP-1D12 reacts with human SLAM-associated protein (SAP, SH2D1A). SAP is a 15-kDa SH2 domain-containing adaptor protein that recruits Fyn tyrosine kinase upon CD150/SLAM receptor ligation, activating downstream signaling.
SAP is expressed in thymocytes, T cells, NK cells, and NKT cells, and plays a role in follicular T helper cell differentiation, NKT cell development, Th1/Th2 differentiation, and regulation of NK/CD8+ T cell cytotoxicity.
Mutations in SAP lead to X-linked lymphoproliferative (XLP) syndrome, associated with impaired humoral immunity and susceptibility to Epstein-Barr virus infection and lymphoma. SAP has also been linked to autoimmune diseases such as systemic lupus erythematosus.

Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested using normal human peripheral blood cells and the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Recommended use: 5 µL (0.03 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

This gene encodes a protein crucial for bidirectional stimulation of T and B cells. It associates with the signaling lymphocyte-activation molecule (SLAM) to inhibit SHP-2 recruitment and modulate signal transduction in activated immune cells.
Mutations cause X-linked lymphoproliferative syndrome (Duncan disease), resulting in susceptibility to Epstein-Barr virus infection and lymphoma. Multiple transcript variants exist for this gene.
