Product Specific Information
This CXNFT monoclonal antibody reacts to mouse NOS2 (inducible NOS, iNOS). iNOS catalyzes nitric oxide formation from L-arginine via an NADPH- and oxygen-dependent mechanism. NOS2 expression is induced by bacterial endotoxins and inflammatory cytokines such as IFN-γ and TNF-α, and is found in M1 macrophages and myeloid-derived suppressor cells.

- Tested Applications: Intracellular staining of mouse thioglycolate-elicited peritoneal exudate cells using Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00)
- Use ≤ 0.125 µg/test (100 µL final volume)
- Cell number: 10⁵–10⁸ cells/test (empirically determined)
- Carefully titrate for optimal performance
Brilliant Violet™ 786 (BV786) is a tandem dye emitting at 786 nm, optimized for cytometers equipped with a 405 nm violet laser.
Important Notes:
- Use Super Bright or Brilliant Stain Buffer to minimize non-specific polymer interactions.
- Protect vial and stained samples from light (photo-induced oxidation sensitivity).
- Fixation: Compatible with IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in dark.
- Not compatible with methanol-based fixation.
- Excitation: 407 nm; Emission: 786 nm; Laser: Violet Laser.

Target Information
iNOS (Inducible Nitric Oxide Synthase)
iNOS produces nitric oxide (NO), a gaseous free radical mediating vasorelaxation, neurotransmission, and cytotoxicity. It is inducible by lipopolysaccharides and cytokines (IFN, TNF, IL-1, IL-2). Found in macrophages, hepatocytes, synoviocytes, and smooth muscle cells. iNOS activity increases after cytokine induction and remains elevated for extended periods. Dysfunction is associated with achalasia and impotence.
