Product Specific Information

The monoclonal antibody SolA15 recognizes mouse and rat Ki-67, a 300 kDa nuclear protein expressed during all active phases of the cell cycle (G1, S, G2, and M), but absent in resting cells (G0). Ki-67 serves as a marker for determining the growth fraction of cell populations and is used in tumor cell studies to evaluate proliferation index.

This antibody also recognizes human, non-human primate, and canine Ki-67.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining of mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended use ≤1.0 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize concentration for best performance.

Fluorochrome Information:
Brilliant Violet™ 786 (BV786) emits at 786 nm and is excited by a 405 nm violet laser. Ensure the cytometer is compatible with this fluorochrome.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. BV786 is not compatible with methanol-based fixation.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle, but absent in G0. It associates with euchromatin and heterochromatin regions and is used as a proliferation marker in neoplastic and neuronal studies. Ki-67 index correlates with tumor grade and proliferation rate, providing valuable prognostic information.
