Product Specific Information

The PK136 monoclonal antibody recognizes mouse NK1.1, an antigen expressed by natural killer cells and a subset of T cells in NK1.1-positive mouse strains such as C57BL and NZB. Strains like BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1; for these, use monoclonal antibody DX5.

Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 shows coexpression of both markers by most cells, with small populations of DX5⁺PK136⁻ and DX5⁻PK136⁺ cells.

Applications Reported: Flow cytometric analysis
Applications Tested: ≤1.0 µg per test (100 µL final volume). Cell numbers may range from 10⁵–10⁸ per test. Titrate for optimal performance.

Brilliant Violet™ 786 (BV786) is a tandem dye emitting at 786 nm, suitable for cytometers with a violet (405 nm) laser. Check instrument compatibility before use.

Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions when using multiple polymer dyes.

Light Sensitivity: Protect vial and samples from light to prevent photo-induced oxidation.

Fixation Compatibility:
Samples can be stored up to 3 days in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) at 4°C in the dark. BV786 is not compatible with methanol-based fixation.

Target Information

NK1.1 (KLRB1, NKR-P1A, CD161) is a C-type lectin-like receptor expressed by NK cells and certain T cell subsets. It mediates cytotoxicity and cytokine secretion upon immune stimulation. The KLRB1 protein features an extracellular lectin domain, transmembrane region, and cytoplasmic domain, functioning as a type II membrane protein. It recognizes Lectin Like Transcript-1 (LLT1) as its ligand.
