Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Alpha actinins belong to the spectrin gene superfamily, a diverse group of cytoskeletal proteins including alpha and beta spectrins and dystrophins.
Alpha actinin is an actin-binding protein with multiple roles in different cell types.

• In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, binding actin to the membrane.
• In skeletal, cardiac, and smooth muscle cells, isoforms are localized to the Z-disc and analogous dense bodies, anchoring myofibrillar actin filaments.
