Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human TPO. Anti-Human TPO-specific antibody was purified by affinity chromatography and then biotinylated.
Sandwich ELISA:
To detect hTPO by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human TPO (500-P49) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant hTPO.
Western Blot:
To detect hTPO by Western Blot, use at 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for Recombinant hTPO is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information
Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor)
TPO is responsible for the generation of platelets through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor MPL (CD110) initiates a signaling cascade via JAK/STAT, MAPK, and PI3K/AKT pathways, leading to proliferation and differentiation of megakaryocytes and platelet production (thrombopoiesis). TPO is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.
