Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human TPO. Anti-Human TPO-specific antibody was purified by affinity chromatography employing an immobilized Human TPO matrix.

Sandwich ELISA:
To detect Human TPO by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. When used with PeproTech Biotinylated Anti-Human TPO (500-P49Bt) as a detection antibody, it enables detection of at least 0.2–0.4 ng/well of Recombinant Human TPO.

Western Blot:
To detect Human TPO by Western Blot analysis, use at 0.1–0.2 µg/mL. With compatible secondary reagents, the detection limit for Recombinant Human TPO is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor) regulates platelet generation through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor MPL (CD110) initiates JAK/STAT, MAPK, and PI3K/AKT signaling pathways, leading to proliferation and differentiation of megakaryocytes and platelet production (thrombopoiesis). Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.
