Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays a key role in lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested

• Pre-diluted and validated for flow cytometry using normal human peripheral blood cells
• Recommended usage: 5 µL (0.8 µg) per test (100 µL final volume)
• Cell count range: 10⁵–10⁸ cells/test

Important Usage Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, constituting ~10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues.
CD45 regulates lymphocyte development and antigen signaling by modulating Src-family kinases and JAK kinases.
Multiple isoforms (e.g., CD45RA) arise from alternative splicing and differ in extracellular domains.
CD45 also influences cell survival, integrin-mediated signaling, and apoptosis regulation.
