Description:
This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (Fc gamma RIII), which exists as two isoforms: FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, and NK cells
• FcγRIIIB: Detected on neutrophils (GPI-anchored form)

Binding of IgG to CD16 triggers ADCC, phagocytosis, cytokine release, and antigen presentation.
3G8 recognizes the same epitope as CB16 but distinct from B73.1.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL/test (1.0 µg/test).
A test = antibody amount for 100 µL final volume, with 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Always use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor Conjugates:
Based on Phiton technology with engineered fluorescent signatures minimizing spillover and spectral spread.
Excitation: 496 nm; Emission: 511 nm; Laser: 488 nm (Blue)

Encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance and cell-mediated cytotoxicity.

  • FCGR3A: NK cells (membrane glycoprotein, transmembrane-anchored)
  • FCGR3B: Neutrophils (PI-linked receptor)
• Clinical relevance:
  Mutations linked to Immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.

• Variants: Alternative splicing yields multiple isoforms.
