Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45, also known as Leukocyte Common Antigen (LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. It plays an important role in lymphocyte activation through tyrosine phosphatase activity.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.8 µg) per test (100 µL final volume).

Usage Notes:

• Cell count range: 10⁵–10⁸ cells/test
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails before mixing with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures with reduced spillover and spread.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells. It comprises about 10% of all surface proteins in lymphocytes and is absent in non-hematopoietic tissues. CD45 regulates Src-family kinases, JAK kinase signaling, and integrin-mediated pathways, playing a crucial role in lymphocyte development, activation, and survival.
