Target Information

RPS6KA5 (MSK1) is a serine/threonine protein kinase that plays a key role in regulating various cellular processes. It is required for mitogen- or stress-induced phosphorylation of transcription factors such as CREB1 and ATF1, and for the regulation of RELA, STAT3, and ETV1/ER81. MSK1 contributes to gene activation via histone phosphorylation and regulates inflammatory gene expression. It phosphorylates CREB1 and ATF1 in response to stimuli such as UV-C irradiation, EGF, and anisomycin. Additionally, MSK1 controls RELA transcriptional activity in response to TNF and glucocorticoids, interacts with NR3C1 (glucocorticoid receptor) in the cytoplasm, and represses inflammatory gene expression. In skeletal myoblasts, it is required for RELA phosphorylation at Ser-276 during oxidative stress, and in erythropoietin-stimulated cells, it mediates Ser-727 phosphorylation of STAT3.
