Target Information
ICAD is the inhibitor of caspase-3-activated DNase (CAD), controlling nuclear apoptosis.
It has two isoforms:
- ICAD-L/DFF45 (45 kDa) – functional isoform
- ICAD-S/DFF35 (35 kDa) – non-functional isoform
ICAD is cleaved during apoptosis, allowing CAD to fragment nuclear DNA.
In non-apoptotic cells, CAD is complexed with ICAD until caspase-3 cleaves ICAD at residues 117 and 224.
ICAD also acts as a CAD-specific folding chaperone.
Recent studies suggest ICAD may be a potential target for restoring normal apoptotic signaling in colon and brain cancer cells.
Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
