Product Specific Information
This antibody reacts with an extracellular epitope of CD11a (alpha subunit of human LFA-1), a 170–180 kDa type I transmembrane glycoprotein expressed on B and T lymphocytes, monocytes, macrophages, neutrophils, basophils, and eosinophils.
Functional application:
The antibody directly induces the binding of T cells to purified ICAM-1. Using an in vitro-translated CD11a cDNA deletion series, the MEM-83 activation epitope was mapped to the I domain of the LFA-1 alpha subunit. These studies identified a novel LFA-1 activation epitope mapping to the I domain, which may play a role in regulating LFA-1 binding to ICAM-1.
Target Information
CD11a (LFA-1 alpha, ITGAL), together with CD18, constitutes leukocyte function-associated antigen 1 (LFA-1), an alphaLbeta2 integrin. CD11a is implicated in activation of the LFA-1 complex and plays a central role in leukocyte intercellular adhesion through interactions with ICAM-1, -2, and -3. It also functions in lymphocyte costimulatory signaling.
CD11a is expressed on leukocyte membranes in a low-affinity conformation. Upon stimulation by chemokines or other signals, it converts to a high-affinity conformation that enables tight binding to ICAMs. CD11a is involved in immune cell interactions, tissue-specific localization, cell differentiation, proliferation, and T-cell effector functions.
Blocking CD11a with specific antibodies or small molecules is an important therapeutic approach in inflammatory disease treatment. For example, the humanized anti-CD11a antibody Efalizumab (Raptiva) interferes with T-cell migration to inflammation sites, while simvastatin binding to the CD11a allosteric site leads to immunomodulation and enhanced lymphocytic cholinergic activity.
Two transcript variants encoding different isoforms of CD11a have been identified. Diseases associated with CD11a dysfunction include benign cephalic histiocytosis and leukocyte adhesion deficiency.
